Bacteria can spontaneously generate and release some specific signal molecules, and can sense the changes in their concentration to regulate the group behavior of microorganisms. This regulation system is called quorum sensing (QS). Bacterial quorum sensing plays an important regulatory role in the interaction between bacteria and hosts.
In Streptomyces, the gamma-butyrolactone quorum-sensing signaling system plays an important role in the regulation of secondary metabolic synthesis. Among them, γ-butyrolactone recognition receptor is widely studied as the basic performer of this signaling pathway. However, there are a large number of "suspected" γ-butyrolactone receptor encoding genes in the Streptomyces genome, some of which have been shown to be unable to bind γ-butyrolactone, so they are called pseudo-γ-butyric acid Lactone receptor protein.
In the early stage, the research work of Tan Huarong's laboratory and Yang Keqian's laboratory of the Institute of Microbiology, Chinese Academy of Sciences confirmed that both JadR2 in Venezuelan zebra mold and ScbR2 in Streptomyces coelicolor are pseudo-γ-butyrolactone receptors. It is specifically combined with endogenous antibiotic molecules and plays an interactive regulation role in different antibiotic synthesis pathways.
Recently, Yang Keqian's laboratory further researched the functions of ScbR2 and JadR2 and found that pseudo-gamma-butyrolactone receptors also have brand-new functions, that is, they can regulate the quorum-sensing signaling molecule γ-butyrolactone in Streptomyces Biosynthesis. ScbR2 and JadR2 specifically terminate the synthesis of γ-butyrolactone, cooperate with the real γ-butyrolactone receptor protein, and control the on and off of quorum sensing signals in Streptomyces. This mode of regulation is common in Streptomyces.
The disclosure of new functions of important regulatory proteins in Streptomyces has deepened people's understanding of the microbial quorum sensing signal system, and it has important guiding significance for the rational transformation of Streptomyces and the scientific improvement of antibiotic synthesis.
