Diaminobenzidine (DAB) is the most sensitive and commonly used chromogenic substrate for horseradish peroxidase. The reaction product is a brown precipitate that is insoluble in water, xylene and alcohol. This product adopts special formula, high sensitivity, low background, stable storage and easy to use. It is suitable for the staining and color reaction of Western blot, immunohistochemistry and immunocytochemistry, dot blot and biochip.

usage

Calculate the amount required for this experiment before use (50-100ul per slice), add 1ml of distilled water to add A liquid, 50ul each of liquid B and liquid C, use within 30 minutes after mixing. Control the color development time under the micromirror, the color development time is 3-30 minutes, too long color development time can cause the background to increase, so the color development process should be closely observed (generally 3-10 minutes is ideal), and the background is shallow And when the appropriate color intensity is reached, the color reaction is terminated by rinsing with running water.

Storage: The kit should be stored at -20 ℃, protected from light and sealed.

Precautions:

1. The DAB solution should be stored at a low temperature and sealed. If crystals precipitate out, make sure the crystals are completely dissolved before use;

2. The color-developing working fluid should be used now, and the freshly prepared working fluid should be colorless or light brown. If the color is too dark, do not use it;

3. DAB is unstable at normal temperature. After each use, it should be sealed and put back in the refrigerator in time, so as not to affect the experiment due to DAB decomposition, or to cause environmental pollution due to leakage;

4. DAB has a potential mutagenic effect, and care should be taken when wearing protective equipment.
